Fig. 2. Expression patterns of the novel genes in anterior ectoderm during normal development. XAgr2 expression at the midneurula stage 15 (A), tailbud stages 28 (B), and 32 (C,D: lateral view). At the stage 32, XAgr2 is expressed in the cells surrounding the cement gland and in the otic vesicle (D, arrowhead). Ras-dva expression at the midneurula stage 15 (E), stage 22 (F), and tailbud stage 25 (F,H: lateral view). At stage 25, Ras-dva expression disappears from the center of developing cement gland, remains in the hatching gland, and starts in the lateral parts of neural tube (H, arrowheads). Nlo expression at the midneurula stage 16 (I), tailbud stages 24 (J), and 27 (K,L: lateral view). At stage 24, Nlo is expressed in the hatching gland (J). At stage 27, Nlo is also expressed in the stripe of cells above the cement gland (K,L, arrows). At stage 32, Nlo is expressed in the otic vesicles (L, arrowhead).

Fig. 3. Sections of the midneurula stage embryos stained for XAgr2 (A,C) and Xanf-1 (B,C) expression. (A) A sagittal section of XAgr2 expression at the midneurula stage. XAgr2 expression is located in the superficial layer of the anterior ectoderm. The arrow indicates the dorsal limit of expression. (B) Xanf-1 expression at the midneurula stage. The arrow indicates the limit of expression in superficial cells, while expression in the deep cells extends more anteriorly. (C) Double in situ hybridization for XAgr2 (shown in red) and Xanf-1 (shown in blue) at the midneurula stage. (D) Scheme of the XAgr2 expression region (shown in red) and the Xanf-1 expression region (shown in blue) in the superficial and deep layers of anterior ectoderm at the midneurula stage.

Fig. 4. Transverse sections of embryos stained for XAgr2 (A,B), Ras-dva (C), and Nlo (D–F) expression. (A) A transverse section of the stage 32 embryo; XAgr2 is expressed in the ventromedial part of otic vesicle. (B) XAgr2 is expressed in the notochord. (C) Ras-dva is expressed in the dorsolateral parts of neural tube at the stage 25 (arrows). (D) A transverse section of the stage 32 embryo; Nlo is expressed in the dorsolateral part of otic vesicle. Nlo is expressed by (E) the basal cells of dorsal fin, and (F) by migrating neural crest cells.
